Not recommended for the diagnosis of pertussis due to low sensitivity. CDC first-line tests for pertussis are PCR (0065080) and/or culture (0060117).

Patient Preparation
Nasopharyngeal swab.  
Specimen Preparation
Place swab in Regan-Lowe transport media and incubate for 24-48 hours at 35°C (ARUP supply# 24962). Available online through eSupply using ARUP Connect™ or contact ARUP Client Services at (800) 522-2787. Also acceptable: Swab in Jones Kendrick media or Amies with charcoal media, or swabs in described media sent directly without incubation.
If both culture and DFA are ordered, two swabs or swab plus slides are required.  
Storage/Transport Temperature
Room temperature or refrigerated.  
Unacceptable Conditions
Sputum. Swabs or media other than listed above.  
Source of specimen is preferred.  
Incubated Regan-Lowe: Ambient: 4 days; Refrigerated: 4 days; Frozen: Unacceptable.
Unincubated Regan-Lowe, Amies with charcoal swab, Aspirate or Washing: Ambient: 48 hours; Refrigerated: 48 hours; Frozen: Unacceptable.
Slides: Ambient: 1 week; Refrigerated: 1 week; Frozen: 1 month

Interpretive Data
DFA is not recommended for diagnosing infections caused by B. pertussis. Refer instead to Bordetella pertussis Culture (ARUP test code 0060117), or Bordetella pertussis/parapertussis by PCR (ARUP test code 0065080) for amplified DNA testing of respiratory specimens.  
A negative stain result does not exclude the possibility of infection. False-negative results may occur due to sampling errors or a low number of organisms in the specimen.
